摘要
【目的】观察SD大鼠大脑中动脉闭塞模型(middle cerebral artery occlusion,MCAO)急性期热休克蛋白70(heat shock response 70,Hsp70)和抗凋亡蛋白Bcl-2(B cell lymphoma/leukemia 2)随时间变化的动态表达,探讨其变化的规律,为临床治疗靶点提供实验依据。【方法】选用健康雄性SD大鼠36只,采用完全随机设计方法分为3组,空白对照组(n=4)、假手术组(n=16)、手术组(n=16)。手术组采用改良线栓法建立急性大脑中动脉闭塞模型,术后0.5 h、2 h、3 h、6 h、12 h、24 h、3 d、7 d共8个时相采血。用ELISA法测定Bcl-2和Hsp70的OD值,绘制标准曲线并计算对应的浓度。【结果】(1)Bcl-2的表达:手术组Bcl-2术后6 h显著升高,12 h到达高峰,3 d时下降接近空白对照组水平。手术组与假手术组在6、12、24 h时间点比较差异有统计学意义(P<0.05);手术组与空白对照组在2 h、3 h、6 h、12 h、24 h、3 d、7 d时间点比较差异有统计学意义(P<0.05);手术组内6、12、24 h与其他时间点比较差异有统计学意义(P<0.05)。(2)Hsp70的表达:手术组Hsp70在术后0.5 h就开始表达,24 h达到高峰,3 d时逐渐下降接近对照组水平。手术组内24 h与各时间点内表达水平比较差异有统计学意义(P<0.05)。【结论】Bcl-2和Hsp70在脑梗死急性期均显著升高,参与神经元缺血缺氧的病理过程,在神经细胞损伤早期发挥重要作用。二者的表达水平呈正相关性。
[Objective] To investigate the dynamic expression of B cell lymphomafleukemia 2(Bcl-2) and heat shock response 70 (Hsp70) in rats after MCAO at different time periods, explore its changing law, and provide an experimental basis to cerebral infarction therapeutic targets. [ Methods] Thirty-six healthy male SD rats were randomly assigned to three groups, control group (n = 4), sham surgery group (n = 16) and surgery group (n = 16). MCAO model was induced by modified Long' s method in surgery group. After surgery took blood sample for 8 times, namely 0.5 hour, 2 hour, 3 hour, 6 hour, 12 hour, 24 hour, 3 day, 7 day eight time points. ELISA method was used to measure the OD values of Bcl-2 and Hsp70 expression levels by standard curve and used analysis of variance to evaluate all data at different time periods and different treatments. [Results] 1)Expression of Bcl-2: After surgery 6 h Bcl-2 significantly elevated, reached peak at 12 h, dropped nearly to the control group level at 3d(P 〈 0.05); Surgery group increased significantly higher than control group at 2 h, 3 h, 6 h, 12 h, 24 h, 3 d, 7 d time point (P〈 0. 05). The 6 h, 12 h, 24 h in surgery group compared with the other time points are significantly different (P 〈 0.05). 2)Expression of Hsp70: After surgery 0.5 h Hsp70 began to express, reached peak at 24 h, declined to nearly to the level of the control group at 3 d. The expression levels at 24 h in surgery group compared with each time point has a significant difference (P 〈 0.05). [Conclusions] Both Bcl-2 and Hsp70 significantly increase in acute cerebral infarction, involved the pathological process in ischemic neuronal hypoxia, play an important role in early nerve cell damage . The expression level of the two parameters there is a certain positive correlation.
