摘要
采用LED红光、LED蓝光以及红蓝光不同配比组合为光源,以荧光灯光质为对照,探讨高山杜鹃组织培养中叶片不定芽再生和试管苗生长对不同LED光质的响应特征。结果显示:红蓝光组合(3∶1)处理与荧光灯对照相比,高山杜鹃叶片不定芽再生率、平均再生芽数均显著增加,而单色红光或单色蓝光处理下高山杜鹃叶片不定芽增殖效果均较差。单色红光处理下高山杜鹃试管苗有一定的徒长现象,株高、叶长和叶宽均最大,茎粗最低,另外,生根率高但根系活力较低;而单色蓝光处理下则相反。在红蓝复色光处理下,高山杜鹃试管苗较健壮且叶片叶绿素含量均高于对照。以上结果说明,红蓝复色光有利于高山杜鹃试管苗的形态建成和叶绿素的合成,是具有明显优势的植物组织培养光源。
The responsive characteristics of leaf regeneration and seedlings growth of Rhododendron in vitro to differ- ent light-emitting diode (LED) light sources were investigated. By using the fluorescent lamp as a control light source, the experiment was carried out under red LED light, blue LED light and three combinations of the two. Compared with control, the shoot regeneration ratio and the numbers of shoots per leaf discs with combinations of red and blue light treatment ( red : blue = 3 : 1 ) were significantly increased, whereas shoot proliferation from leaf treated with red LED light or blue LED light was poor. The greatest stem length, leaf length, leaf width and rooting rate, and the thinnest stem diameter and the poorest root vigor of seedlings were obtained in red LED light treatment, while the blue LED light had the opposite effects. The seedlings exposed to combinations of red and blue LED light showed stronger growth and higher chlorophyll contents than those exposed to fluorescent light. It was concluded that the combinations of red and blue LED light benefited the morphogenesis and chlorophyll biosynthesis of Rhododendron seedlings in vitro, and it could be the light source for plant tissue culture.
