摘要
目的:探讨β-榄香烯体外逆转人肺腺癌细胞PC9耐吉非替尼的作用及其机制。方法:使用噻唑蓝(MTT)比色法测定不同药物处理对耐吉非替尼的肺腺癌细胞株PC9/ZD的抑制率,FCM检测各组细胞的凋亡情况,Western blot检测PC9/ZD各组细胞磷酸化细胞外调节激酶(p-Erk)及磷酸化AKT(p-Akt)的表达水平。结果:MTT实验结果显示PC9/ZD的联合用药组抑制率(41.69±5.06)%和单药组抑制率(38.20±5.54)%分别与吉非替尼对照组比较,差异有统计学意义(P<0.05);流式细胞检测结果显示与吉非替尼对照组比较,联合用药组的凋亡率(15.17±2.01)%与单药组的凋亡率(7.59±1.35)%与吉非替尼对照组(5.63±1.19)%比较作用显著;Western blot检测检测发现,联合用药组的p-Akt和P-Erk蛋白表达量明显减少。结论:β-榄香烯具有逆转PC9/ZD细胞耐吉非替尼的作用,可能与下调耐药细胞内p-Erk、p-Akt表达水平相关。
Objective : To explore the role of elemene in reversing human lung adenocarcinoma cell line PC9 against gefitinib and its possible mechanisms. Methods : Gefitinib resistant cell line PC9/ZD cells were treated with the combination of elemene and gefitinib or separately, and then the cell proliferation inhibition rates were determined by MTT method. The apoptosis rate of PC9/ZD cells was examined by flow cytometry. The expression levels of phosphorylated Akt ( p - Akt) and the phosphorylated extracellular signal -regulated kinase (p -ERK) in PC9/ZD cells were detected by Western blotting. Results : It was demonstrated by MTY that the growth of PC9/ZD cells was inhibited in the combined group and single groups compared with control group ( P 〈 0.05 ). It revealed by flow cytometry that the PC9/ZD cell apoptosis rate was higher in the combined group and single groups compared with control group, and the combined group was the highest. Western blotting revealed that protein levels of p - Erk and p - Akt were inhibited significantly compared with control group. Conclusion: β- elemene can reverse the drug resistance of PC9/ZD,which probably is related with its down - regulation of p - Erk and p - Akt protein expression.
