塞来昔布减轻糖尿病大鼠心肌细胞纤维化的实验研究 被引量:1

摘要 目的探讨塞来昔布对糖尿病心肌病大鼠心肌细胞纤维化的影响。方法将Wistar大鼠30只随机分为对照组(A组)、糖尿病心肌病组(B组)和糖尿病心肌病塞来昔布治疗组(C组)各10只。取大鼠心肌组织,经常规石蜡切片、染色后测定胶原纤维面的密度值;采用RT-PCR法测定TGF-β1mRNA表达情况。结果与A组相比,B组大鼠心肌组织的胶原纤维密度值、TGF-β1mRNA表达明显升高(P<0.05)。C组大鼠的胶原纤维、TGF-β1mRNA表达明显低于A组和B组(均P<0.05)。结论塞来昔布可能通过抑制TGF-β1减轻糖尿病大鼠心肌纤维化。 Objective To investigate the effect of celecoxib on myocardial fibrosis in diabetic rats. Methods Wistar rats were randomly divided into normal control group (A group, n= 10), diabetic myocardial group (B group, n= 10) and diabetic cardiomyopathy celecoxib treatment group (C group, n= 10). The rats were sacrificed and myocardial tissues underwent paraffin and staining tissue sections, the surface density of collagen fibers level was detected. The expression of transforming growth factor-βl (TGF-β1) mRNA was detected by RT-PCR. Results Compared with the normal control group, the collagen fibers of myocardial tissue in diabetic rats, TGF-β1 mRNA expression was significantly increased (P〈0. 05). The collagen fibers in celecoxib treated rats, TGF-β1 mRNA expression was significantly lower than the diabetic myocardial group and normal control group (P〈0. 05). Conclusion Celecoxib can alleviate development of myocardial fibrosis in diabetic rats.
