摘要
目的 考察不同炮制方法对水蛭的氨基酸含量和抗凝血酶活性的影响.方法 采用柱前衍生化高效液相色谱法测定水蛭不同炮制品的游离氨基酸及水解氨基酸的含量;采用凝血酶滴定法测定其抗凝血酶活性.结果 蚂蟥的游离氨基酸含量为0.64%, 水解氨基酸含量为24.7%, 抗凝血酶活性为14.4 U;柳叶蚂蟥1 批的游离氨基酸含量为0.51%, 水解氨基酸含量为35.7%, 抗凝血酶活性为14.9 U;柳叶蚂蟥2 批及其炮制品的游离氨基酸含量为0.68%~0.78%,水解氨基酸含量为33.6%~39.9%,抗凝血酶活性为9.6~15 U.结论 柳叶蚂蟥的游离氨基酸含量及抗凝血酶活性与蚂蟥差别不大,但水解氨基酸含量高于蚂蟥;柳叶蚂蟥经过炮制后,游离氨基酸及水解氨基酸含量变化不大,而高温炮制(砂炒、蜜炙麦麸炒、滑石粉炒)降低了其抗凝血酶活性.
Objective To study the effects of different processing methods on amino acid content and the anti-thrombin activity of Leech. Methods Hydrolytic and free amino acids content in leech were determined by the pre-column derivatization HPLC method. The anti- thrombin activity of Leech was determined by the thrombin titration. Results The content of free amino acid in Whitmania pigra was 0.64%, hydrolytic amino acid was 24.69%, antithrombin activity was 14.4 U. The content of free amino acid in the first batch Whitmania acranulata was 0.51%, hydrolytic amino acid was 35.69%, antithrombin activity was 14.9 U. The content of free amino acid in the second batch Whitmania acranulata and its processed products were 0.68%- 0.78%, hydrolytic amino acid was 33.61%-39.93%, antithrombin activity was 9.6-15 U. Conclusion There are not significant differences between Whitmania pigra and Whitmania acranulata in the content of free amino acids and antithrombin activity, but the hydrolytic amino acid contents in Whitmania acranulata is higher than that in Whitmania pigra. There are no obvious differences in the contents of free amino acids and hydrolytic amino acids before and after processing, while an- tithrombin activity in Leech was reduced by high temperature processing.
