摘要
目的探讨胶质源性神经营养因子(glial cell line-derived neural factor,GDNF)基因修饰的神经干细胞(neural stem cells,NSCs)移植对大鼠脑缺血后(glial fibrillary acidic protein,GFAP)的表达的影响。方法建立大鼠暂时性大脑中动脉梗塞模型(middle cerebral artery occlusion,MCAO),再灌注3d,并将其随机分为:生理盐水移植组(NS)、神经干细胞移植组(NSCs)和GDNF基因修饰的神经干细胞移植组(GDNF/NSCs)。采用脑立体定位仪将移植入大鼠的右侧脑室。再灌注1、2、3、5、7周的各时间点,处死大鼠,取同侧大脑半球,用Western blotting观察GFAP的表达。结果 GDNF/NSCs组、NSCs组和NS组GFAP蛋白均在2周达到高峰,后逐渐降低。而再灌注1~7周,GDNF/NSCs组的GFAP蛋白显著高于NS组(P〈0.001);GDNF/NSCs组GFAP蛋白显著高于NSCs组(P〈0.01)。再灌注1~5周,NSCs组GFAP蛋白显著高于NS组(P〈0.001)。结论 GDNF/NSCs移植治疗大鼠缺血再灌注的神经保护作用,其机制可能与促进GFAP的表达有关。
Objective To investigate the effect of transplantating neural stem cells(neural stem cells,NSCs) modified by glial cell line-derived neural factor(GDNF) gene on expression of glial fibrillary acidic protein(GFAP) in rats with cerebral ischemia.Methods The cerebral ischemia model was performed by occluding middle cerebral artery for 2 hour and reperfusiong.At 3 days of reperfusion,saline,NSCs and GDNF / NSCs were administratied by a stereotaxic frame into right lateral ventricle.Then the rats were survived for 1,2,3,5 and 7W,respectively and followed to be sacrificed.Western blot was used to detect GFAP expression.Results GFAP expressions of GDNF / NSCs,NSCs and NS groups peaked at 2 weeks,then gradually decreased.At reperfusion 1 to 7 week,GFAP expressions of the GDNF / NSCs group were significantly higher than the NS group(P〈0.001).At reperfusion 1 to 5 week,NSCs group GFAP expression was significantly higher than NS group(P〈0.001).Moreover,GFAP expressions of GDNF / NSCs group was significantly higher than NSCs group(P〈0.01) at given time.Conclusion Transplantating GDNF / NSCs may increase GFAP expression in rats following ischemia,which may be one of the mechanisms that GDNF / NSCs tansplantaion provides the neruoprotection.
