牦牛HSPA1A和HSPA2基因组织分布的检测 被引量:1

摘要 旨在建立一种检测牦牛HSP70家族应激型HSPA1A和HSPA2基因的实时荧光定量PCR方法。采用普通PCR,结合实时荧光定量PCR方法,检测正常情况下应激型HSPA1A和HSPA2基因在牦牛各组织中的表达分布。结果显示,HSPA1A基因在公牦牛与母牦牛中的表达基本一致,所有组织中均少量表达,肺中最低,肌肉相对表达较高;HSPA2基因在公牦牛与母牦牛中的表达也基本一致,所有组织均少量表达,肺中最低,脑、睾丸和卵巢相对表达较高。该研究结果提示正常情况下HSPA2基因可能在牦牛生殖繁育上发挥一定作用,为进一步研究HSPA1A基因和HSPA2基因在牦牛对环境的适应性方面提供基础资料和研究方法。 This study was to establish a real-time PCR assay for detection of stress type HSPA1Aand HSPA2 genes of HSP70 family of yaks,and further detection of expresssion and distribution of these two genes by using PCR combined with real-time PCR.The results showed that the detection resulsts of PCR and real time PCR were basically similar.The expression levels of HSPA1A gene in male yaks and female yaks were both similar,all of the tissues were very low in expression,lung is the lowest and the muscle was relative high.The expression levels of HSPA2 gene in male yaks and female yaks were also basically similar,all of the tissues were very low in expression lung is the lowest and the brain,testis,ovary was relative high.The results suggest that HSPA2 gene may play a role in reproductive breeding of yaks in normal condition.These results provide the basis study on the environmental adaptability of yaks.
