红花黄色素对大鼠气道黏液高分泌的治疗作用 被引量:2

摘要 目的讨论红花黄色素在气道慢性炎症黏液高分泌机制中的作用。方法以注射用红花黄色素为可变因素设为治疗组,应用中性粒细胞弹力酶雾化吸入法建立大鼠气道黏液高分泌机制病理模型,注射用水设为对照组,用RT-PCR、ELISA法检测各组气道上皮黏蛋白的表达情况,以western blot和放射活性法分别测定蛋白激酶C(PKC)的蛋白水平以及PKC活性。结果红花黄色素组的黏蛋白mRNA转录和蛋白表达水平均较对照组明显降低,治疗组组织中蛋白激酶C(PKC)蛋白水平及活性也有明显下降。结论红花黄色素可通过抑制PKC活性而抑制大鼠气道黏液高分泌。 Objeetive To investigate the efects of Chinese traditional medicine safflower yellow on mucin MUC5AC expression in mucous hyperseretion rats. Methods Used a rat model of neutrophil elastase (NE)-induced airway inflanunation, experiment group was treated with safflower yellow solution, and isotonic Na chloride was used in control group. MUC5AC mRNA and protein were detected by RT-PCR and ELISA, respectively. Protein kinase C(PKC)protein expression was assayed by western blot, activity of PKC was detected by radiation method. Results NE increased mucin expression and PKC activity in rats(P〈0.01), compared with normal control group. When treated with safflower yellow, MUC5AC mRNA expression, MUC5AC protein production and PKC activity were decreased obviously(P〈0.05), compared with NE stimulated group. Conclusion Safflower yellow can reduce mucous hypersecretion in rats airway epithelial through inhibition of PKC.
