摘要
采用DNA-SRAP分子标记技术和NTSYS类平均法对29份枸杞种质材料的亲缘关系进行了初步分析。结果表明,所用的5对引物共扩增出19条带,其中多态性条带为14条,多态性比率达76.68%。聚类分析表明,以0.74,0.83和0.91的相似系数可将全部供试种质分为3种、5种和15种类群。因此认为,在分子水平上,枸杞种质材料间的遗传多样性非常丰富;黑果枸杞与供试种质的遗传距离较远;SRAP分子技术可应用于枸杞种质鉴定和亲缘关系分析。
The genetic relationship of 29 wolfberry germplasms were analyzed by the technology of genomic DNA-SRAP markers and the NTSYS average similarity cluster analysis .The results showed that 19 bands were amplified u-sing 5 pairs of primers , 14 of which were polymorphic bands , and polymorphic rate was 76.68%.The cluster analy-sis showed that all germplasms were classified into 3 groups, 5 groups and 15 groups according to different coefficient of 0.74 , 0.83 , and 0.91 .The genetic diversity of wolfberry germplasms is very ample at the molecular level .Lycium ruthenicum is farther from other wolfberry germplasms in the genetic distance .It is showed that the technology of SRAP markers can be applied to identification of germplasm and analysis of genetic relationship in wolfberry .
