摘要
以甘蔗新品种桂糖02—1156为材料,生产甘蔗脱毒苗。为了筛选其最佳培养基配方,在培养基内添加PP333、6-BA和NAA并采用均匀设计法设计试验方案,结果采用二次回归法进行分析。试验结果表明,PP333取lmg/L,6-BA取2mg/L,其脱毒苗株型好,增殖率高,预测值为5.974,经验证试验其增殖率为6.045,与预测结果基本符合。因此,将均匀设计法应用到甘蔗组培快繁中筛选培养基配方是一种高效,准确率高的方法。
New sugarcane variety Guitang 02-1156 was used for micropropagation of virus-free seedlings. PP333, 6-BA and NAA were added in the culture medium, and the uniform design method was taken in the test. The results were analyzed by the quadratic regression method. Test results showed that MS+ PP333 1 mg/L+ 6- BA 2 mg/L was the most suitable medium composition with good plant-type and high proliferation rate, the predicted value was 5.974, by verified its proliferation rate was 6.045, and the results were basically in agreement with predictions. So the uniform design was highly efficient and accurate, it could be applied to the rapid propagation of screening culture medium composition in sugarcane.
