荧光实时定量PCR方法在肺癌相关基因检测中的应用 被引量:1

摘要 目的研究实时定量荧光PCR在非小细胞肺癌患者肿瘤组织、正常组织及外周血中RRM1、ERCC1表达水平检测的应用价值。方法构建目的基因质粒标准品,运用PCR仪进行SYBR荧光实时定量分析,比较其表达水平。结果两者在外周血中表达水平与TNM分期、是否淋巴转移等有关;肿瘤组织和外周血中的表达具有一致性。结论荧光实时定量PCR具有良好的特异性和灵敏度,费用低,操作简便,可应用于临床检验工作。 Objective To study the detection value of real-time quantitative PCR on the expression of RRMI and ERCC1 in patients with non-small cell lung cancer tumor tissue, normal tissue and peripheral blood. Methods The expression of RRMI and ERCCI was measured by SYBR real time analysis through PCR. Results Their ex- pression in the peripheral blood was associated with TNM stage and lymph node metastasis. The expression in tumor tissue and peripheral blood was consistent. Conclusion Real-time PCR has the advantages of high specificity and sensitivity, low cost, and easy operation.
