摘要
目的:研究表面修饰聚对苯二甲酸乙二醇酯(PET)材料对骨髓间充质干细胞增殖、迁移能力的影响及机制。方法:取SD大鼠进行骨髓间充质干细胞培养,并参考Atthoff的方法进行PET材料表面修饰;表面修饰的PET材料与骨髓间充质干细胞复合培养为观察组,常规PET材料与骨髓间充质干细胞复合培养为对照组,通过MTT的方法检测细胞活力,划痕试验检测细胞迁移能力,Western-blot的方法检测基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)、半胱氨酸蛋白酶-3(Caspase-3)、半胱氨酸蛋白酶-9(Caspase-9)的蛋白水平。结果:观察组细胞的细胞活力、迁移能力均高于对照组,差异有统计学意义(P<0.05);观察组细胞的MMP-2、MMP-9含量高于对照组,Caspase-3、caspase-9含量低于对照组,差异有统计学意义(P<0.05)。结论:表面修饰的PET人工韧带体与间充质干细胞复合培养有助于增强细胞的迁移和增殖能力,MMP-2、MMP-9、Caspase-3、Caspase-9是其可能的分子机制。
Objective:To investigate the effects of surface modification PET material on proliferation and migration of bone marrow mesenchymal stem cells and its mechanism.Methods:Bone marrow mesenchymal stem cells were cultured from SD rats,PET material were modified by the ways of Atthoff. Co-culture of surface modified PET material and mesenchymal stem cells was observation group,co-culture of conventional PET material and mesenchymal stem cells was control group.Then cell vitality was detected by MTT,migration were detected by wound-healing assay and protein level of MMP-2,MMP-9, Caspase-3,Caspase-9were detected by Western-blot.Results:(1)Vitality(198.4±29.5 vs.100±17.2) and migration(235.2±41.4 vs.100.0±41.4)of observation group were higher than those of control group showing significant different(P0.05);(2)MMP-2and MMP-9levels of observation group were higher than those of control group,but Caspase-3and caspase-9levels of observation group were significant lower than those of control group(P0.05).Conclusion:Co-culture of surface modified PET material and mesenchymal stem cells can enhance the migration and proliferation ability of cell and MMP-2, MMP-9,Caspase-3,Caspase-9are possible molecular mechanism.
