摘要
目的 观察抗微管新药紫杉醇对B细胞淋巴瘤细胞株BJAB是否具有凋亡诱导作用 ,并进一步研究bcl 2基因家族在此过程中的作用。方法 将不同浓度的紫杉醇作用于BJAB细胞 ,观察其作用的时间效应及剂量效应 ;在光镜和电镜下观察其形态变化 ;用流式细胞仪分析细胞DNA含量的改变并做DNA片段分析 ;用免疫组织化学及半定量RT PCR法观察在紫杉醇作用过程中bcl 2基因家族的蛋白及mRNA的变化。结果 紫杉醇能抑制BJAB细胞生长 ,抑制作用首先表现为G2 /M期阻滞 ,一定时间后出现细胞凋亡 ,并显示剂量和时间效应。在这一过程中 ,bcl 2转录及蛋白表达下降 ,并出现bcl xs的转录。结论 紫杉醇可诱导BJAB细胞凋亡 ,这为其应用于B细胞淋巴瘤的治疗提供依据。bcl 2和bcl
Objective To study the apoptosis inducing effect of taxol on BJAB B cell lymphoma cell line and the role of bcl 2 gene family. Methods BJAB cells were treated with taxol and examined for morphologic changes under light and electron microscopes. Flow cytometry assays and DNA gel electrophoresis were used to demonstrate changes in nuclear DNA contents and DNA fragmentations. Changes in bcl 2 gene family at mRNA and protein levels were studied by immunohistochemistry and semi quantitative RT PCR technique.Results Taxol inhibited growth of BJAB cells. The cells initially showed G 2/M arrest, followed by apoptosis, in a time and dose dependent manner. The expression of bcl 2 at mRNA and protion level was decreased while that of bcl xs transcription was increased after taxol treatment.Conclusion Taxol induces apoptosis of BJAB cells. Bcl 2 and bcl xs are involved in the regulation of taxol induced apoptosis.
