人类细胞凋亡芯片筛选骨肉瘤凋亡相关蛋白

摘要 目的通过人类细胞凋亡芯片筛选骨肉瘤中表达异常的凋亡相关蛋白,为近一步深入功能机制研究奠定基础。方法应用人类细胞凋亡芯片,筛选骨肉瘤组织及癌旁组织中表达有差异的凋亡相关蛋白。应用ImageJ1.42图片处理软件求出每个孔的灰度值,进行统计分析。结果凋亡相关蛋白Bcl-2、cIAP-2、Clusterin、HSP27、Survivin在骨肉瘤组织中和癌旁组织中都有表达,且其在骨肉瘤组织中的表达水平大于癌旁组织(P<0.05)。结论凋亡蛋白Bcl-2、cIAP-2、Clusterin、HSP27、Survivin在骨肉瘤组织中表达上调,可能参与骨肉瘤的发病机制。 Objective To detect apoptosis-related proteins abnormally expressed in osteosarcoma.Methods We extracted proteins in osteosarcoma tissue and adjacent tissue,and used the Human Apoptosis Array select apoptosis protein of abnormal expression.Image J 1.42,a image processing software was used to calculate gray value of each hole for statistical analysis.Results Bcl-2、cIAP-2、Clusterin、HSP27、Survivin were expressed in osteosarcoma tissue and adjacent tissue.Gray value of osteosarcoma tissue was greater than adjacent tissue(P0.05).Conclusion Bcl-2、 cIAP-2、Clusterin、HSP27、Survivin expression increases in osteosarcoma tissue,and they may be involved in the pathogenesis of osteosarcoma.
