摘要
以玉米谷蛋白为原料,研究了不同水解条件下Alcalase 2.4L蛋白酶水解玉米谷蛋白制备的酶解产物的还原能力。在单因素试验的基础上,考察加酶量、温度、时间、pH值和底物浓度5个因素对水解产物还原能力的影响,并通过响应面分析法优化酶解条件,得到的最佳工艺条件为:加酶量3.35%,水解温度56.4℃,时间4.15h,pH8.3,底物浓度0.01g/ml。在此条件下,测得玉米谷蛋白酶解产物的还原能力为0.673。
With corn glutelin as materials, the reducing capacity of corn glutelin hydrolysate prepared by Alcalase 2.4 1. under different hydrolysis conditions was studied. On the basis of single factor experiments, the effects of enzyme amount, tempera ture, hydrolysis time, pH and substrate concentration on reducing capacity of the hydrolysate were studied, and the enzymatic hydrolysis conditions were optimized by response surface methodology. The optimum conditions were as follows: enzyme a- mount 3.35%, temperature 56.4℃, hydrolysis time 4.15 h, pH 8.3 and substrate concentration 0.01 g/ml. Under the opti- mum conditions, the reducing capacity of corn glutelin hydrolysate was 0. 673.
