摘要
【目的】基于尿动力学探讨缩泉丸及其提取物缩尿的作用机制。【方法】采用水提、醇沉、水蒸汽蒸馏法提取缩泉丸。选用SD大鼠以D-半乳糖诱导亚急性衰老模型,随机分为模型对照组、缩泉丸组、水提物组、挥发油组、醇沉物组。造模时间持续8周,第3周开始同时灌胃给予相应药物,空白对照组、模型对照组给予等容积蒸馏水,给药周期为8周。每隔1周对动物进行24 h尿量测定,末次给药后,收集24 h尿液测定水通道蛋白2(AQP2)的含量,最后1周进行5 h水负荷实验。末次给药后对各组大鼠进行尿动力学检测。【结果】与空白对照组比较,模型对照组24 h尿量、5 h水负荷尿量均显著增加,尿中AQP2含量显著减少,膀胱漏尿点压(BLPP)、腹压漏尿点压(ALPP)、最大膀胱排尿压(MVP)显著降低,最大尿道压(MUP)、尿道闭合压(MUCP)显著增加,膀胱顺应性(BC)显著增加(P<0.05或P<0.01)。与模型对照组比较,缩泉丸组及醇沉物组尿量显著降低,缩泉丸组、水提物组、醇沉物组尿中AQP2含量均显著增加,缩泉丸组及水提物组BLPP、MVP均显著升高,BC显著降低(P<0.05或P<0.01);醇沉物组BLPP、ALPP均显著升高(P<0.05或P<0.01)。挥发油组尿动力学各项指标与模型组比较差异均无统计学意义(P>0.05)。【结论】缩泉丸可改善亚急性衰老大鼠的尿动力学异常,3种提取物中醇沉物的整体效果较好,可能通过其多糖成分改善人体整体功能而起作用。
Objective To observe the urodynamic mechanism of Suoquan Pills (SP) for reducing urination. Methods SP were extracted with water, ethanol and steam distillation, respectively. After subacute aging rats model was established by injection of D-galactose, all the rats were divided into model control group, SP group, SP water extract group, SP ethanol extract group and SP volatile oil group. Additionally, blank control group was set up. From the 3rd week of modeling, medication groups were given the corresponding medicine, and model control group and blank control group were given distilled water, administration lasting for 8 weeks. During the modeling period, 24-hour urine volume was tested every other week, 24-hour urine was collected after the last medication for the examination of aquaporin (AQP) 2 content, and 5-hour water loading test was carried out during the last week. At the end of experiment, urodynamic parameters were determined. Results In the model control group, 24-hour urine volume and urine volume after 5-hour water loading were significantly increased, urine AQP2 content, bladder leak point pressure (BLPP) , abdominal leak point pressure (ALPP) and maximum voiding pressure (MVP) were significantly decreased, maximum urethral pressure (MUP) and maximum urethral closure pressure (MUCP) were significantly increased, and bladder compliance (BC) was also increased (P〈0.05 or P〈0.01 compared with blank control group), which indicated that the model has been established successfully. Compared with the model control group, urine volume in SP group and SP ethanol extract group was decreased (P〈0.05 or P〈0.01), AQP2 content was increased in SP group, SP water extract group and SP ethanol extract group, BLPP and MVP were increased and BC was decreased in SP group and SP water extract group (P〈0.05 or P〈0.01), and BLPP and ALPP were increased in SP ethanol extract group (P〈0.05 or P〈0.01) . Urodynamic indexes in SP volatile oil group did not differ from the model control group (P〉0.05) . Conclusion SP can distinctly improve urodynamic disorders in subacute aging rats. The ethanol extract of SP shows better effect on improving general body function than water extract and volatile oil, which is probably related with the polysaccharides of SP.
