白芍总苷对大鼠腹膜纤维化转化生长因子-β1/Smad7信号转导通路的影响 被引量:7

Effect of Total Glucosides of Paeony on Transforming Growth Factor β1/Smad7 Signal Transduction Pathway in Peritoneal Fibrosis Rats

摘要 【目的】探讨白芍总苷对大鼠腹膜纤维化转化生长因子-β1(TGF-β1)及其信号转导分子7(Smad7)的干预作用及其机制。【方法】采用42.5 g/L含糖透析液+脂多糖法复制腹膜纤维化模型。将SD大鼠随机分成4组:空白组,模型组,白芍总苷高、低剂量组。第30天后,测定各组腹膜功能,取腹膜组织进行Masson染色观察腹膜病理学变化,采用实时荧光聚合酶链反应(Real-time PCR)方法检测各组腹膜TGF-β1 mRNA、Smad7 mRNA的表达。【结果】模型组腹膜厚度显著增加,腹膜超滤量显著降低(均P<0.05)。白芍总苷高、低剂量组对上述病变均有显著改善作用(P<0.05)。模型组腹膜TGF-β1 mRNA、Smad7 mRNA表达水平显著高于空白组(P<0.05或P<0.01),白芍总苷高、低剂量组TGF-β1表达较模型组均显著降低(P<0.05),白芍总苷高剂量组抑制性信号蛋白Smad7则显著升高(P<0.05)。【结论】白芍总苷抑制腹膜组织TGF-β1 mRNA表达、上调Smad7 mRNA表达可能是其抑制腹膜纤维化的机制之一。 Objective To investigate the effect of total glucosides of paeony (TGP) on transforming growth factor (TGF)-β1/Smad7 signal transdnetion pathway in rats with peritoneal fibrosis and to explore its possible mechanism. Methods The model of peritoneal dialysis fibrosis was established by giving dialysis fluid containing 42.5g/L sugar and lipopolysaccharide. And then the model rats were randomly divided into four groups, blank group, model group, and high- and low-dose TGP groups. After treatment for 30 days, peritoneal function in all of the groups was measured. Peritoneal tissue was stained by Masson for observing the pathological changes. TGF-β1 and Smad7 mRNA expression levels in peritoneal were detected by real-time quantitative polymerase chain reaction (PCR) . Results The thickness of the peritoneum was significantly increased and the peritoneal ultrafiltration was the worst in the model group (P〈0.05), while the above changes were improved in high-and low-dose TGP gr oups (P〈0.05) . TGF-β1 and Smad7 mRNA expression levels in the model group were significantly higher than those in the blank group (P〈0.05 or P〈0.01) . TGF-β1 was significantly lowered in high- and low- dose TGP groups and Smad7 was increased in high-dose TGP group (P〈0.05 compared with those in the model group) . Conclusion TGP can inhibit peritoneal fibrosis induced by peritoneal dialysis and improve peritoneal structures, and one of its possible mechanisms is related with the inhibition of TGF-β1 and the increase of Smad7 mRNA expression in the peritoneal tissue.
