Optimization of Fermentation Culture of Amanita pallidorosea and Their Antifungal Action on Pathogens

摘要 优化玫瑰红鹅膏液体发酵培养条件,并研究其对植物病原菌活性的抑制作用。采用中心组合旋转设计法确定玫瑰红鹅膏液体深层培养菌丝体的最佳条件,采用HPLC和ESI-MS法对其发酵产物中的肽类毒素进行检测。采用生长速率法研究玫瑰红鹅膏子实体粗毒液和发酵产物对香瓜枯萎病菌的抑制作用。当温度25℃、pH 6、转速156r·min-1时,菌丝体重量达到最高,为1.054 g。从发酵产物中分离鉴定出2种化合物:α-AMA和β-AMA。玫瑰红鹅膏子实体粗毒液和发酵产物对香瓜枯萎病菌有很好的抑制作用,玫瑰红鹅膏子实体粗毒液浓度为200 g·mL-1时,抑制率达最高为42.47%。玫瑰红鹅膏发酵产物中检测出2种肽类毒素:α-AMA和β-AMA;玫瑰红鹅膏子实体能有效的抑制香瓜枯萎病菌的活性,具有开发利用价值。 To optimize fermentation culture of A. pallidorosea and to study their antifungal action on pathogens, optimum condi-tions of fermentation culture of A. pallidorosea were investigated by using central composite rotatable design, and we used HPLC and ESI-MS to detect main peptide toxins from fermentation products. The antifungal action of the crude extraction of A. paUidorosea and fermentation products on Fusarium oxysporum were studied by using growth rate method. The results showed that the weight of mycelium was 1. 054 g when temperature was 25℃, pH was 6 and speed was 156 r·min^-1. Two peptide toxins of α-AMA and β-AMA were got. The crude extraction of A. pallidorosea and fermentation products had significant antifungal ac- tion on Fusarium oxysporum. The highest inhibition ratio on Fusarium oxysporum was 42. 47% when concentration of crude ex-traction of A. pallidorosea was 200 g· mL^-1. Two peptide toxins from fermentation products were detected. A. pallidorosea could effectively inhibit the activity of Fusarium oxysporum, which had the value of development and utilization.
