摘要
目的:观察辛伐他汀对2型糖尿病大鼠心肌Toll样受体4(TLR4)表达的影响。方法:40只雄性SD大鼠随机分为对照组(n=10)和实验组(n=30)。其中对照组给予普通饲料,实验组用高糖高脂饲料喂养。6周后测定空腹血糖>7.8 mmol/L且伴有胰岛素敏感指数降低的大鼠作为2型糖尿病大鼠模型,共26只,将其随机分为糖尿病组(n=13)和辛伐他汀组(n=13)。干预治疗8周。留取心脏组织做光镜观察,分别采用免疫组织化学染色和逆转录多聚酶链反应法(RT-PCR)检测各组大鼠心肌TLR4蛋白和信使核糖核酸(mRNA)的表达。结果:与对照组比较,光镜下可见糖尿病组心肌细胞明显水肿,并可见大量炎细胞浸润,辛伐他汀组心肌组织损伤较糖尿病组明显减轻。免疫组织化学染色和RT-PCR显示,糖尿病组大鼠TLR4蛋白和mRNA表达分别为0.6157±0.0233和1.43±0.16,明显高于对照组(0.3210±0.0151和0.42±0.06,P<0.05);辛伐他汀组大鼠TLR4蛋白和mRNA表达分别是0.4302±0.0224和0.73±0.13,低于糖尿病组(P<0.05),但仍高于对照组(P<0.05),差异均有统计学意义。结论 :辛伐他汀从蛋白和mRNA水平下调2型糖尿病大鼠心肌TLR4的表达、降低炎症反应,进而发挥对心脏的保护作用。
Objective: To observe the effect of simvastatin on reducing the expression of myocardial Toll like receptor 4 (TLR4) in type 2 diabetic rats. Methods: A total of 40 male SD rats were divided into 2 groups, Control group, n=10, me rats had normal feed and Experimental group, n=30, the rats received hyperglycemia and hypelipaemia feed for 6 weeks. There were 26 experimental rats with fasting blood glucose 〉7.8 mmol/L defined as diabetic rats and divided into 2 groups, Diabetic group, the rats received the normal saline, and Simvastatin group, the rats received simvastatin 20mg/(kg·d) for 8 weeks, n=13 in each group. The myocardial tissue was examined by optical microscopy, TLR4 protein and mRNA expressions were measured by immuno histochemistry and RT-PCR respectively in different groups. Results: Compared with Control group, the myocardial tissue in Diabetic group showed obvious edema and inflammatory cell infiltration, those damages were much less in Simvastatin group. The immuno-histochemistry and RT- PCR presented that TLR4 protein and mRNA expressions in Diabetic group (0.6157 ± 0.0233) and (1.43 ± 0.16) were significantly higher than those in Control group (0.3210 ± 0.0151) and (0.42 ± 0.06) all P〈0.05. The TLR4 protein and mRNA expressions in Simvastatin group (0.4302 ± 0.0224) and (0.73 ± 0.13) were lower than Diabetic group but higher than Control group, all P〈0.05. Conclusion: Simvastatin may reduce the expressions of TLR4 protein and mRNA and decrease the inflammatory reaction therefore protect the cardiac tissue in experimental rats.
