绵羊肺腺瘤病毒SD株的鉴定及分子特征分析 被引量:2

摘要 采用组织病理学和分子生物学方法对山东某地区疑似绵羊肺腺瘤病毒感染的肺组织进行观察,H.E.染色可见肺泡上皮细胞增生,呈乳头状突入肺泡腔,周围肺泡气肿,肺间质可见淋巴细胞浸润,符合绵羊肺腺瘤病毒感染的病理学特征;PCR检测及序列分析表明,从该肺组织中扩增出了绵羊肺腺瘤特异性env基因,全长2 249bp,与内源性绵羊肺腺瘤病毒南非株enJS56A1(AF153615)的核苷酸同源性为98.9%,且处于同一个进化分支,而与外源性绵羊肺腺瘤病毒美国株JSRv21(AF105220)的核苷酸同源性仅为81%,提示SD株属于内源性肺腺瘤病毒。本次研究为绵羊肺腺瘤病毒的分子流行病学研究积累了资料,也为实验室确诊该病毒病提供了方法参考。 The study aims to analyze the lung tissue of sheep samples infected by the suspected SPA virus in a certain area of Shandong province by histopathology and molecular biology. By HE staining, these clinical lesions can be seen: papillary alveolus ep- ithelial hyperplasia protruding into the alveolar cavity, with alveolar ectasia surrounding it and interstitial lymphocytic infiltrates, which correspond to the histopathological features of sheep infected by SPA virus. PCR test and sequential analysis show that SPA specific env gene obtained from the lung tissue is 2 249bp long, and its nucieotide homology with enJS56Al(AF153615) of endoge- nous SPA virus is up to 98.9% and they belong to the same evolutionary branch;however, its nucieotide homology with JSRV21 (AF105220) of exogenous SPA virus is only 81%, which shows that SD strain belongs to endogenous SPA virus. This study accumu- lates useful data for the future research on the molecular epidemiology of SPA virus and provides referential method for the identifica- tion of this virus in laboratory.
