摘要
目的研究血红素加氧酶-1(HO—1)对ALI大鼠的保护作用和对肺组织水通道蛋白1(AQP-1)、AQP-5表达的影响及可能的调节机制。方法30只Wistar大鼠随机分成3组,即生理盐水组、ALI组和血红素组。尾静脉注射脂多糖后观察4h,光镜下观察肺组织病理改变程度,测定肺组织湿/干重比值,检测大鼠血清中IL-8及环磷酸腺苷(cAMP)的含量;免疫组化方法观察肺组织AQP-1、AQP-5及H0-1的表达。结果①与生理盐水组相比,ALI组肺组织湿/干重比值显著升高(P〈0.001),血清IL8含量升高(P〈0.05);血红素组肺组织湿/干重比值及IL-8含量均低于ALI组(P〈0.05),肺损伤炎症减轻。②血红素组AQP-1、AQP-5的表达均显著高于ALI组(Pd0.001)。③ALI组大鼠血清cAMP含量与生理盐水组比较差异没有统计学意义,血红素组cAMP含量较ALI组增加(P〈0.05)。结论HO-1可以上调AQP-1、AQP-5的表达,减轻ALI时的肺水肿,可能是通过抑制炎症因子IL-8的表达实现的。cAMP可能参与了AI。I时AQP-1与AQP-5的调节。
Objective To research the protective effects of heme oxygenase-1 (HO-1) on ALI rats, the influence of HO-1 on the expression of aquaporin-1 (AQP-1) and AQP-5 in lung tissue, and the possible mechanism. Methods Thirty Wistar rats were randomly divided into three groups:normal saline treated group,ALI model group, and heroin induced group. The rats were observed for four hours after injecting lipopolysaccharide in tail vein, the optical microscope was used to observe the pathological changes of lung tissue,wet/dry ratio in lung tissue was measured, and the content of IL-8 and cAMP in rat serum was determined. The immunohistochemical method was used to detect the expressions of AQP-1, AQP-5,and HO-1 in lung tissue. Results ①Compared with normal saline treated group, the wet/dry ratio increased significantly in ALl model group ( P 〈0. 001), and the IL-8 level in serum in ALI model group was also higher than that in normal saline treated group ( P〈0.05). The wet/dry ratio and IL-8 level in serum in hemin induced group were lower than those in ALI model group ( P 〈0.05),and the inflammation of lung injury was reduced. ②The expressions of AQP 1 and AQP-5 in hemin induced group were significantly higher than those in ALl model group (P 〈 0. 001). ③There was no statistical significance on the content of cAMP between ALI model group and normal saline treated group. The content of cAMP in hemin induced group was higher than that in AI.I model group ( P〈 0.05). Conclusions HO-1 may upregulate the expressions of AQP-1 and AQP-5, reduce the degrees of pulmonary edema in ALI rats possibly through inhibiting the expression of inflammatory factor IL-8 .cAMP may be involved in the regulation of AQP 1 and AQP-5 in ALI .
