摘要
目的 :建立测定异维A酸血药浓度的方法。方法 :在避光条件下 ,血浆标本加磷酸盐缓冲液后用乙醚提取 ,吹干后以二氯甲烷 -冰醋酸 (99 5∶0 5 )为流动相 ,经过YWGSi6 0柱分离后在紫外 36 5nm处检测。结果 :血药浓度标准曲线在 10~ 5 0 0ng·mL-1,r=0 9999,最低检测限为 10ng mL-1,高、中、低 3种浓度平均回收率为 81 6 2 %~ 8 8 99%。日内和日间平均RSD小于 0 6 1%和 8 2 0 %。结论
Objective:To establish a HPLC method for determining isotretinoin in human plasma.Method:Under yellow light,the plasma was extracted with ether after being put in phosphate-buffered saline.The isotretinoin was eluted with dichoromethane-acetic acid (99 5∶0 5) on YWG Si60 column and detected at 365 nm.Results:Calibration curve is linear from 10~500 ng mL -1 ,r=0 999 9 The limit of detection is 10 ng·mL -1 .The range of mean recovery was 81 62%~88 99%.The RSD of intraday and interday were less than 0 61% and 8 20% respectively.Conclusion:The present study prevides a reliable and simple quantitative method for pharmacokinetic studies of isotretinoin.
