摘要
目的:观察蒿芩清胆汤对幽门螺杆菌相关性胃炎(HAG)小鼠血清白细胞介素-8(IL-8)水平及胃黏膜IL-8、核转录因子κB p65(NF-κB p65)的影响,探讨其对胃黏膜损伤修复的作用机制。方法:按体重、性别随机抽取10只小鼠作为空白对照组,其他小鼠采用幽门螺杆菌(HP)标准菌株(SS1)布氏肉汤液灌胃造模。造模成功后,50只模型小鼠按性别、体重随机分为5组,每组10只,即为模型组、丽珠维三联组(0.25 g·kg-1)、蒿芩清胆汤低、中、高剂量组(4,8,16 g·kg-1)。灌胃容积10mL·kg-1,空白对照组、模型组灌服等体积生理盐水,1次/d。2周后处死动物,观察各组小鼠胃黏膜病理学改变,酶联免疫法(ELISA)检测血清IL-8含量、免疫组化法检测胃黏膜IL-8,NF-κB p65的表达。结果:与模型组比较,蒿芩清胆汤能显著降低HAG小鼠胃黏膜炎症面积和平均灰度(P<0.01);蒿芩清胆汤明显降低HAG小鼠血清IL-8含量(P<0.01);蒿芩清胆汤能明显降低HAG小鼠胃黏膜IL-8,NF-κB p65的表达(P<0.05)。结论:蒿芩清胆汤对HAG小鼠胃黏膜损伤有明显修复作用,其机制可能与其抑制NF-κB p65的激活、下调IL-8的过度表达有关。
Objective: To investigate the effect of Haoqin Qingdan decoction on the content of interleukin 8(IL-8) in serum and expression of IL-8, neclear factor-κB p65(NF-κB p65) in Gastric mucosa in Helicobacter pylori-associated gastritis(HAG) mice and explore its therapeutic mechanism for Helicobacter pylori(HP) associated gastritis. Method: HP standard strains (SS1) brinell broth liquid was used to establish the HAG mice model. Sixty healthy BALB/c mice were randomly devided into six group (n=10, each), normal control group(given with equal NS), model group (given with equal NS), livzon weisanlian group(0.25 g·kg-1), Haoqin Qindan decoction (4,8,16 g·kg-1) group. The mice in every group were administrated with drugs by oral gavage once a day. All mice were killed two weeks later, the pathological changes of each group were observed by the microscope, the contents of IL-8 in serum were detected by enzyme-linked immunoassay and the expression of IL-8, NF-κB p65 in gastric mucosa tissue were detected by immunohistochemistry. Result: Compared with model group, Haoqin Qingdan decoction significantly reduced the inflammation area and average blackness of gastric mucosa in HAG mice (P〈0.01);the expression of IL-8 in serum of Haoqin Qingdan decoction groups were significantly lower than that in model group (P〈0.01);the expression of IL-8, NF-κB p65 in gastric mucosa of Haoqin Qingdan decoction groups were significantly lower than that in model group (P〈0.05). Conclusion: Haoqin Qingdan decoction can plays a significant role in the repair of gastric mucosal injury in HAG mice, its therapeutic mechanism may be related to inhibiting the activation of NF-κB p65 and degrading the expression of IL-8 in the gastric mucosa and serum.
