摘要
目的研究葛根总黄酮经鼻给药对利血平致偏头痛模型动物的主要治疗作用及其机制。方法采用皮下注射利血平制备大鼠偏头痛模型,实验设阴性对照组、模型组、麦角胺咖啡因组和葛根总黄酮组。除麦角胺咖啡因组给药1次外,其余各组共给药3次,每次间隔20 min。各组首次给药后60 min取血与脑组织,观察给药后大鼠脑内去甲肾上腺素、多巴胺、高香草酸、5-羟色胺(5-HT)、5-羟吲哚乙酸等神经递质水平的变化及大鼠血液中一氧化氮合酶(NOS)水平的变化。结果模型组动物脑内多巴胺、5-HT水平显著降低,血液中的NOS水平明显升高;滴鼻给予不同剂量的葛根总黄酮能明显提高利血平致偏头痛模型动物脑内去甲肾上腺素、多巴胺的水平,大鼠血液中的NOS水平显著降低。结论葛根总黄酮滴鼻给药能明显改善利血平致偏头痛的症状。
AIM To observe the protective effect and mechanism of total flavone from Pueraria by nasal drug delivery on reserpine-inducing migraine rats. METHODS The subcutaneous injection of reserpine was performed on rat to make the model for migraine. Rats were divided into negative control group, model group, ergotamine caffeine group and three total flavone from Ptteraria groups. Rats of ergotamine caffeine group were treated by a single oral gavage. Rats of other groups were treated three times at an interval of twenty minutes. Rats of negative control or model group were treated with water of equal volume. Blood and brains of rats were collected in sixty minutes af- ter first administration. The effect of total flavone from Pueraria was observed on noradrenalin, dopamine, 5- hydroxytryptamine (5-HT) , homovanillic acid, 5-hydroxyindole acetic acid in brain and nitric oxide synthase in blood. RESULTS The levels of dopamine, 5-HT in brain of migraine rats induced by reserpine significantly de- creased, but the level of nitric oxide synthase (NOS) in blood significantly increased. The levels of noradrenalin, dopamine in brain of migraine rats induced by reserpine significantly increased after treatment with different dosage total flavone from Pueraria compared with model group. The level of NOS in blood reduced significantly in total flavone from Pueraria group and ergotamine caffeine group. CONCLUSION Total flavone from Pueraria could reduce the symptoms of migraine induced by reserpine in rats.
