摘要
目的探讨CXCL12-CXCR4趋化因子轴与卵巢癌发病的关系。方法将转染质粒SKOV3/CXCR4、转染载体SKOV3/neg及未转染的SKOV3三种卵巢癌细胞进行体外培养,建立荷人卵巢癌细胞裸鼠腹腔移植瘤模型,观察各组裸鼠自然死亡后腹腔成瘤数、体积、腹水量和存活期的变化。结果 3组裸鼠生存时间、腹腔移植瘤的个数和重量比较差异有统计学意义(P<0.05),CXCL12浓度越高,受试裸鼠腹腔移植瘤个数越多,重量越大,生存时间越短。结论 CXCL12-CXCR4趋化因子轴与卵巢癌的发生有一定的相关性,可能是其发病的一个重要原因,在一定程度上也反映了病情的轻重。
Objective To investigate the correlation between chemokine CXCL12-CXCR4 and ovarian cancer. Methods The three kinds of ovarian cancer cells including SKOV3 transfected with plasmid, SKOV3 transfected with vector and SKOV3 without transfection were cultured in vitro. The transplantation tumor models in nude mice were established by intraperitoneal injection with tumor ceils, after they died naturally, the tumor amount in abdominal cavity, tumor size, ascites volume and mean survival time were observed in the 3 groups. Results There were significant differences in the mean survival time, the number and weight of transplantation tumor among three groups ( P 〈 0.05). The higher the concentration of CXCL 12,the more transplantation tumor,the more weight the tumors and the shorter the survival time would be. Conclusion The chemokine CXCL12-CXCR4 is related to pathogenesis of ovarian cancer at some extent, which may be one of pathogenic reasons and reflects severe degree of the disease at some extent.
