摘要
目的优选佛手的最佳醇提工艺,以期对大生产提供试验依据。方法以佛手代表性成分———橙皮苷为考察指标,对HPLC外标法测定橙皮苷含量进行了方法学考察;比较了20%、30%、40%、50%、60%、70%、80%等7种不同浓度的乙醇以及浸渍提取、超声提取、加热回流提取等3种不同提取方法对佛手提取效果的影响;然后采用L9(34)正交试验法,考察乙醇用量、提取时间及提取次数对提取效果的影响。结果提取次数与乙醇用量对考察指标具有显著性影响。佛手的最佳醇提工艺为加40倍药材量的30%乙醇,加热回流提取3次,每次0.5 h。结论优化后的橙皮苷含量测定方法稳定,准确可靠,为佛手质量的评价提供了依据。
Objective To optimize the extraction technology of Citrus lnedica L. var. sarcodactylis Swingle with alcohol. Methods The extraction effects were investigated by different concentrations (20% ,30% ,40% ,50% ,60% ,70% ,80% ) of ethanol,as well as diver- sified extraction processes (such as immersion extraction,ultrasonic extraction, reflLtxing extraction) , additionally,the orthogonal test of L~ (34) was employed to study the effect of quantities of ethanol,extraction time and extraction fi-equeneies and the results were deter- mined by HPLC with Hesperidin of Citrus lnediea L. vat'. sareodaetylis Swingle as the observation index. Results The extraction fi'e- queney and quantity of ethanol played a prominent role on the extraction effect,while the optilnal condition was 40-fold 30% (v/v) eth- anol the amount of medicinal material at a reflttx for 30 luln and extracted three times. Conclusion The optimized extraction technology shows stabilization and accuracy,which can be used to evaluate Citrus lnediea L. var. sareodactylis Swingle.
