摘要
目的建立枳根口服液中总多糖的含量测定方法。方法供试品经60%乙醇醇沉去杂质、氯仿—甲醇(2∶1)溶液脱脂、Sevag混和液(氯仿∶正丁醇为5∶1)除蛋白,90%乙醇醇沉得总多糖,以苯酚—硫酸法,在490 nm波长处测定总多糖含量。结果回归方程为A=0.001 8X-0.007 4(r=0.999 1),葡萄糖的检测质量浓度在26.636~266.36 mg·L-1范围内与吸光度呈良好的线性关系,测定方法经精密度、稳定性、重现性、加样回收试验验证,证明方法科学、可行。结论该方法简便、准确、重现性好,可用于枳根口服液中总多糖的含量测定。
Objective To establish a meflmd for content determination of pdysacc^haride in Zhigen oral solution. Methods The sam- pies were prepared by water extraction-alcohol precipitation,derailed by trichloromethane-lnethanol(2:1). The protein was relnoved by trichlorolnethane-n-butanol(5:1) ,the method of content determination of pdysaceharide was by phenol-sulfuric acid method and UV photometry at 490nin. Results The regression equation was A 0. 001 8X- 0. 007 4. The related coefficient is 0. 999 1. The linear relationship of polysaccharide' concentration was at the range of 26. 636 - 266.36 mg·L-1. Good linearity, high accuracy and precision, wide range and long durability were obtained in this way. Conclusions This method is sensitive, accurate and rapid,which can be used for quality contrd of coment determination of polysaccharide in Zhigen oral solution.
