摘要
目的建立荧光原位杂交技术(fluorescent in situ hybridization,FISH)检测PDGFRB基因重排的方法。方法依据PDGFRB断裂重排方式,设计、制备双色荧光探针。以人外周血培养淋巴细胞为检测对象评价PDGFRB基因重排检测探针的灵敏度和特异性,建立阴性阈值。以白血病患者的骨髓和外周血样本为对象进行PDGFRB重排检测性能评价。结果本研究建立的FISH检测方法在人外周血培养淋巴细胞检测中特异性和灵敏度均达到100%,在对28例样本检测中,筛查出1例PDGFRB FISH阳性样本。结论本研究制备的双色荧光探针可用于PDGFRB基因重排检测。
Objective To develop an assay for PDGFRB rearrangement detection based on FISH technology. Methods According to PDGFRB rearrangement manner, dual-color FISH probes were designed and prepared. Human peripheral blood lymphocytes as testing object to evaluate the sensitivity and specificity of the probe that detected PDGFRB gene rearrangement, and established the negative threshold. Results Both specificity and sensitivity of this FISH assay reached 100% in peripheral blood lymphocytes. In 28 cases of samples, one cases of PDGFRB positive samples was screened out by FISH. Conclusion The dual-color fluorescent probes could be prepared for PDGFRB gene rearrangement detection.
