摘要
目的 观察肿瘤性巨噬细胞(TAMs)在肿瘤微环境中对人结肠癌细胞侵袭性的影响.方法 模拟肿瘤微环境,分别通过Transwell共培养小室将TAMs(1×109/L)和结肠癌细胞Lovo(1×109/L)及SW480(1×109/L)进行共培养24h,收集TAMs 24 h的上清液加入Lovo(l×109/L)及SW480(1×109/L)中培养24 h,再经Transwell侵袭实验检测两株细胞的侵袭性.结果 肿瘤细胞共培养后均提高了侵袭性,其中Lovo[(344±220个比(230±12)个,P<0.05],SW480[(303±23)个比(211±18)个,P<0.05];而加入TAMs上清的肿瘤细胞侵袭性未获得明显的增强,其中Lovo[(257±22)个比(221±17)个,P>0.05];SW480[(233±11)个比(209±19)个,P>0.05].结论 TAMs在肿瘤微环境能够与结肠癌细胞相互作用,从而提高结肠癌细胞侵袭性.
Objective To investigate the influence of tumor associated macrophages (TAMs) on invasion of colorectal cancer cells (CRCs).Methods To imitate tumor microenvironment,Transwell chamber was used to co-culture TAMs (1 × 109/L) with CRCs Lovo (1 × 109/L) and SW480 cells (1 ×109/L),or supernatants collected from TAMs at 24 h and then added into Lovo (1 × 109/L) and SW480 (1 × 109/L) cells for another 24 h.The invasion ability of Lovo and SW480 cells was tested by using Transwell invasion assay.Results The invasion ability of both the two co-cultured cancer cells was significantly increased (for SW480,303 ± 23 vs.211 ± 18,P 〈 0.05 ; and for Lovo,344 ± 22 vs.230 ± 12,P 〈 0.05).The invasion ability of the cancer cells incubated with conditioned medium was not significantly enhanced (for Lovo,257 ± 22 vs.221 ± 17,P 〉 0.05 ; and for SW480,233 ± 11 vs.209 ± 19,P 〉 0.05).Conclusion TAMs could improve CRCs invasion through the interaction in the tumor microencironment.
