摘要
目的探讨秦皮甲素对小鼠肺癌移植瘤的影响及作用机制。方法分组对比观察秦皮甲素对小鼠Lewis肺癌移植瘤的影响,瘤组织称重计算抑瘤率,免疫印迹法检测秦皮甲素对凋亡基因Bax、Bcl-2的影响。流式细胞仪检测瘤细胞增殖周期及凋亡,JC-1染色检测线粒体膜电位。结果秦皮甲素可抑制小鼠肺癌移植瘤的生长和瘤细胞的增殖周期,降低线粒体膜电位,在上调Bax蛋白表达的同时下调Bcl-2蛋白表达,且呈剂量依赖性。结论秦皮甲素对小鼠肺癌移植瘤有显著的抑制作用。使肿瘤细胞阻滞于S期,并通过线粒体途径诱导凋亡。
ABSTRACT: AIM To explore the effect and mechanism of aesculin on mouse xenotransplanted Lewis lung canc- er. METHODS Mouse Lewis lung cancer xenograft model was used to study the effect of aeseulin on tumor growth. The tumor tissue was weighted to caIculate the antitumor rate. Influence of aesculin on apoptotic genes Bax, Bcl-2 was determined by Western blot. Distribution of tumor cell cycle and apoptotic cell death were analyzed by flow cytometry and mitochondrial membrane potential JC-1 staining. RESULTS Aescufin inhibited mouse lung tumor growth and tumor cell proliferation cycle and reduced the mitochondrial membrane potential. Aesculin pro- moted Bax protein expression and at the same time lowered Bel-2 protein expression in a dose dependent manner. CONCLUSION Aesculin has a significant inhibition against the growth of mouse xenotransplanted Leise lung cancer, whose cycles are arrested at S phase. Aesculin could induce apoptosis through the mitochondrial pathway.
