SAHA联合GCV对EBV阳性鼻咽癌细胞的细胞毒作用

摘要 目的探讨伏立诺他(SAHA)联合GCV对EBV阳性鼻咽癌细胞的细胞毒作用,为临床化疗提供理论依据。方法在EBV阳性鼻咽癌细胞C666和EBV阴性鼻咽癌细胞NP69中,通过MTT检测SAHA联合GCV的细胞毒性;利用Western-Blot检测SAHA诱导C666细胞裂解期蛋白EA-D的表达;利用流式细胞仪检测SAHA及其联合GCV诱导细胞凋亡情况。结果 MTT结果显示SAHA联合GCV能增强GCV的细胞毒性,随着时间延长SAHA诱导C666 E-D表达增多;SAHA及其联合GCV诱导细胞凋亡没有统计学差异(P>0.05)。结论 SAHA联合GCV增强了GCV的细胞毒性,此协同作用可能与SAHA诱导EBV阳性鼻咽癌细胞进入裂解期有关。 Objective To explored the cytotoxicity effect of SAHA and GCV in EBV positive nasopharyngeal caJrcinoma (NPC) cells to provide theory evidence for clinical chemotherapy. Methods The cytotoxicity of SAHA and GCV in EBV positive NPC cells C666 and negative cells NP69 were detected by MTF. EBV lytie cycle protein EA-D expression was detected by Western-Blot. The apoptosis of C666 induced by SAHA or combined with GCV was detected by flow cytometry. Results The results showed the cytotoxicity of GCV could be enhanced by SAHA in C666 cells not in NP69 ceils by MTY, and EA-D protein expression was increasing in C666 cells However, there is no significant statistically differences between the apoptos/i/s combined with GCV (P 〉 0.05 ). Conclusion The cytotoxicity effect of^CV due to EBV entering the lyric cycle induced by SAHA. induced by SAHA as time getting longer. results of C666 induced by SAHA and could be enhanced by SAHA, which may
