超高效液相色谱法快速测定膨化食品中7种人工合成色素 被引量:17

Determination of seven synthetic pigments in puffed foods by ultra performance liquid chromatography

摘要 目的建立了一种使用超高效液相色谱快速同时测定膨化食品中7种人工合成色素的检测方法。方法将样品粉碎均匀,加入乙醇+氨水+水(7+2+1)的提取液直接超声提取样品中色素,采用Waters ACQUITY UPLC BEH C18(2.1 mm×50 mm,1.7μm)色谱柱,以甲醇和0.02 mol/L的乙酸铵水溶液为流动相,梯度洗脱,流速为0.25 ml/min,柱温25℃,二极管阵列检测器检测,检测波长为254 nm和627 nm,外标法峰面积定量。结果线性范围柠檬黄1μg/ml^200μg/ml,新红1μg/ml^200μg/ml,苋菜红1μg/ml^200μg/ml,胭脂红2μg/ml^200μg/ml,日落黄1μg/ml^200μg/ml,诱惑红2μg/ml^200μg/ml,亮蓝2μg/ml^200μg/ml,线性关系良好,相关系数0.9996~0.9999,平均回收率分别为87%~105%,相对标准偏差0.5%~3.1%,检出限分别为0.10 mg/kg^0.30 mg/kg。结论该本方法前处理简单、快速、准确,适用于食品中7种人工合成色素的定量分析。 Objective To establish an ultra high performance liquid chromatography(UPLC) method for determination of seven synthetic pigments in foods. Methods The sample was extracted by ethanol -ammonia hydroxide -water(7:2:1 ) , separated on a Waters ACQUITY UPLC BEH C18 column(2.1 mm × 50 mm, 1.7 μm) by gradient elution using a mobile phase of metha- nol and 0.02 mol/L ammonium acetate at flow rate of 0.25 ml/min and the column temperature at 25 ℃, then detected by di- ode array detector at 254 nm and 627 nm, and quantified with external standard method. Results The calibration curves showed a good linearity with correlation coefficients from 0.9996 to 0.9999, and the linear range was 1 μg/ml - 200 μg/ml for lemon yellow, new red, amaranth and sunset yellow, and 2 μg/ml-200 μg/ml for carmine, allura red and light blue. The av- erage recovery rate ranged form 87% to 105% with relative standard deviation from 0.5% to 3.1%. The detection limit was from 0.10 mg/kg to 0.30 mg/kg. Conclusion The method is simple, rapid and accurate, it can be used for quantitative anal- ysis of synthetic pigments in food at the same time.
