摘要
目的了解兔跟腱在愈合过程中相关生长因子表达的变化。方法采用45只新西兰大白兔,建立兔跟腱横断伤后修复的模型。于术前、术后3天、7天、2周、3周、4周、6周、8周天切取手术侧及对侧相应部位跟腱。检测正常跟腱、损伤侧跟腱(实验组)和损伤对侧跟腱(对照组)中的相关生长因子(TGF-β1、bFGF、VEGF、PDGF和IGF-1)表达的变化。采用双抗夹心ELISA法,分析不同修复阶段跟腱中的生长因子的含量。结果实验组的生长因子分别在不同的时相较对照组有不同程度的增高及逐渐恢复正常的变化。结论兔跟腱损伤各个阶段各个生长因子的表达都有不同程度的增高,随着修复的完成,其浓度趋于伤前水平。
Objective Study the rabbit tendon correlated growth factor’s expression in repairing. Methods 45 New Zealand rabbits were divided into eight groups randomally, which are normal group, 3-day group, 7-day group, 2-week group, 3-week group, 4-week group, 6-week group, 8-week group, 5 rabbits per group, another 5 rabbits reserve for death and wound infection. Each left posterior limb are control group, right posterior limb are experimental group which are operated. Normal group is the normal control, which are not operated. The materials of other groups are drawn partly from posterior limbs at corresponding phase and detected correlated factors, such as TGF-β1, bFGF, VEGF, PDGF, IGF-1 and so on. Detecting the content of growth factors with ELISA reagent-box. Results The growth factors in experimental group are differential higuer than the control one, and then getting right gradually. Conclusion The growth factors are differential increasing after injury, and then getting right gradually with the end of repairing.
