寒饮蕴肺证COPD大鼠模型肺组织AQP1、Muc5ac表达的动态研究 被引量:11

摘要 目的:动态研究寒饮蕴肺证慢性阻塞性肺疾病(COPD)大鼠肺组织中水通道蛋白1(AQPl)、黏蛋白5ac(Muc5ac)的表达与证候之间的关系,以及温阳化饮方对以上指标的影响。方法:130只SPF级雄性Wistar大鼠随机分为对照组、自然演变组和温阳化饮方治疗组,采用免疫组化法动态检测(观察60d,每隔10d检测1次)肺组织中AQPl、Muc5ac的表达。结果:在撤除造模因素后60d内,自然演变组肺组织中AQPl表达持续降低,Muc5ac表达持续升高。温阳化饮方治疗组肺组织AQPl表达升高,Muc5ac表达降低,治疗10--40d后疗效显著,40d后疗效不明显。结论:肺组织AQPl表达的降低,Muc5ac表达的升高与证候的演变关系密切,温阳化饮方能够上调肺组织中AQPl的表达和下调Muc5ac的表达,对COPD寒饮蕴肺证有明显的治疗作用。但治疗存在时效性。 Objective:To dynamically study the relationship between protein AQP1 and Muc5ac expression and the syndrome,and the effects of Warming Yang and Dispersing Fluid Formula on chronic obstructive pulmonary disease(COPD) with syndrome of accumulation of cold fluid in the lung. Methods :130 SPF male Wistar rats were randomly divided into control group,natural evolution group and Warming Yang and Dis- persing Fluid Formula-treated group,using the immunohistochemical method dynamically detect(observation of 60 days,testing every 10 days) the expression of AQP1 and Muc5ac in lung tissue. Results:After the re- moval of building factors within 60 days,natural evolution group continued to reduce AQP1 expression in lung tissue,and the expression of Muc5ac continued to rise. Warming Yang and Dispersing Fluid Formu- la-treated group lung tissue AQP1 expression increases,and reduced expression of Muc5ac. Effect is obvious after 10-40 days,and not obvious after 40 days. Conclusion:AQP1 expression decrease and Muc5ac expres- sion increase of lung tissue are closely associated with the evolution of the syndrome. Warming Yang and Dispersing Fluid Formula can increase the expression of AQP1 in lung tissue and lower the expression of Muc5ac,which may have potential therapeutic role in COPD with syndrome of accumulation of cold fluid,but the treatment has timeliness.
