摘要
以革兰氏阳性菌金黄色葡萄球菌标准菌株为对照,以大肠杆菌标准菌株为总大肠菌群的代表菌,通过大量的染色试验,初步探讨菌种菌龄、涂菌状态、碘液媒染时间和酒精脱色时间4个关键控制点对水质检验中总大肠菌群革兰氏染色效果的影响,结果表明菌种菌龄22-26 h,涂菌状态用接种环边缘挑取少量典型菌分散涂布,碘液媒染时间20-60 s,酒精脱色时间30-50 s为最优方案。
The Gram-positive bacterium standard strain of Staphylococcus aureus is used as the comparison and the standard strain of E. coli is used as the representative bacteria of the total coliform in this paper. Through a large number of the staining tests, the influences of the bacteria strain age, the coating mode of the bacteria, the iodine solution mordant agent time and the alcohol-decolorization time as the four critical control points on the result of total coliforms Gram staining on water quality testing are investigated preliminarily. The results show that the excellent Gram staining program is as follows: the bacteria strain age should be between 22 and 26 hours. The coating mode of the bacteria should be a small amount of scattered typical bacteria at the edge of the inoculating loop. The iodine solution mordant agent time should be between 20 and 60 seconds and the alcohol-decolorization time should be between 30 and 50 seconds.
