摘要
目的:研究地塞米松合用中药淫羊藿女贞子对哮喘大鼠糖皮质激素受体(GR)亚型及转录因子的影响。方法:实验大鼠随机分为7组:正常对照(N)组、哮喘模型(A)组、激素干预(GC)组、淫羊藿女贞子(YN)组、淫羊藿女贞子合激素干预Ⅰ组(YN+GC组Ⅰ)、淫羊藿女贞子合激素干预Ⅱ组(YN+GC组Ⅱ)、淫羊藿女贞子合激素干预Ⅲ组(YN+GC组Ⅲ)。哮喘大鼠在激素干预不同阶段合用淫羊藿女贞子,激素撤停2周后处死,检测血清GR亚型含量、肺组织GR亚型mRNA和54种转录因子蛋白表达。结果:中药合激素干预的3个组均显著抑制血清中GRβ含量,但上调肺GRβmRNA表达。GC组肺组织有43种转录因子表达较哮喘组下调,仅有1种上调;与GC组比较,中药合激素干预的3个组分别有49种、45种、46种转录因子表达上调。结论:淫羊藿女贞子合用地塞米松的协同抗炎及抑制激素的副作用与影响GR亚型及上调肺转录因子的表达有关。
Objective: To observe the effects of the compatibility of Epimedium and Fructus Ligustrilucidi on subtypes of glucocorticoid receptor (GR) and transcriptional factor (TF) of asthmatic rats with glucocorficoid (GC) intervention. Methods: Dexamethasone was injected into asthmatic model rats with the oral gavage of Epimedium and Fructus Ligustrilucidi at different treatment stages of GC withdraw. After GC withdraw 2 weeks, the rats were sacrificed. The contents of serum subtypes of GR and the expression of subtypes of GR mRNA and 54 TFs protein of lung tissue were observed. Results: The contents of serum GR Ⅰ in group I, I and III of Chinese medicine and GC significantly decreased, while the expression of GRI3 mRNA in lung tissue significantly increased. 43 TFs showed increased activities, only I showed decreased activities, compared with asthma group. Gl:oup I, II and III of Chinese medicine and GC had 49, 45 and 46 kinds of TF expression significantly increased respectively, compared with group GC. Conclusion: The mechanism of synergistic anti-inflammatory and inhibiting adverse effects of Epimedium and Fructus Ligustrilucidi combined Dexamethasone is related to GR subtypes and increasing the expression of TFs in lung tissues.
