摘要
目的:研究银杏达莫注射液(Ginkgo leaf extract and dipyridamole injection,GLED)降低家兔动脉血压的作用及机制。方法 :采用颈总动脉插管法测动脉血压。经静脉累积注射GLED(0.35、0.7、1.4、2.8、5.6、11.2 mg/kg)观察血压的变化。并分为:左旋硝基精氨酸(10 mg/kg)、异搏定(10μg/kg)、普萘洛尔(0.5 mg/kg)、吲哚美辛(0.5 mg/kg)、缬沙坦(1.5 mg/kg)5组,观察它们对GLED(5.6 mg/kg)降压作用的影响。结果 :GLED能显著降低家兔动脉血压,并具有量效依赖性(收缩压:r=0.76,t=8.4,P<0.01;舒张压:r=0.76,t=6.5,P<0.01)。静脉注射10 mg/kg左旋硝基精氨酸后,GLED(5.6 mg/kg)的降压作用明显减弱,异搏定、普萘洛尔、吲哚美辛和缬沙坦对GLED(5.6 mg/kg)的降压作用无明显影响。结论 :GLED能显著降低家兔动脉血压,其机制可能与内皮释放的NO有关。
Objective: To study Ginkgo Leaf Extract and Dipyridamole Injection ( GLED ) reducing arterial blood pressure of rabbits in vivo and the possible mechanism. Methods: Rabbits common carotid artery was intubated and arterial blood pressure in vivo was recorded. GLED with cumulative concentrations of 0.35,0.7,1.4,2.8,5.6,11.2 mg/kg was injected into vein and the changes of arterial blood pressure were observed. In addition, rabbits were divided into five groups, including 10 mg/kg N co - nitro - L - arginine (L- NN A ), 10 f.L g/kg verapamil (Vera), 0.5 mg/kg propranolol (Prop), 0.5 mg/kg indomethacin (Indo), 1.5 mg/kg valsartanal (Vals handom. 5.6 mg/kg GLED was injected into vein respectively and the changes of arterial blood pressure were observed. Results: GLED had a significant concentration-dependent negative effect on arterial blood pressure in vivo (the systolic pressure: r=0.76, t=8.4, P〈O.OI ; the diastolic pressure: r=0.76, t=6.5, P〈O.OI ). The effect of the decreasing arterial blood pressure of GLED (5.6 mg/ kg) was significantly reduced by injection into vein with 10 mg/kg L-NNA, but there was no obvious effect by treatments respectively with Vera, Prop, Indo or Vals. Conclusion: GLED can reduce arterial blood pressure significantly in vivo. The mechanism of reducing arterial blood pressure may be related to NO releasing from endothelial cells.
