氟对大鼠肾脏组织中caspase-3和caspase-9蛋白表达影响 被引量:5

摘要 目的探讨氟对大鼠肾脏组织细胞凋亡过程中caspase-3和caspase-9蛋白表达的影响。方法 48只SD大鼠分为对照组和低、中、高染氟组(氟化钠50、100、200 mg/L),每组12只,120 d后测尿氟含量,处死大鼠,称体重及肾脏脏器重量,计算肾脏脏器系数,肾病理切片观察及caspase-3和caspase-9免疫组化测定。结果与对照组比较,高氟组大鼠体重降低(P<0.01),中氟组肾脏脏器系数降低(P<0.05),各染氟组大鼠尿氟含量均升高(P<0.01),且尿氟含量与染氟浓度呈剂量-效应关系(r=0.925,P<0.01);染氟组肾小球体型变大,肾小管排列结构紊乱、疏松,肾小球内皮细胞和肾小管上皮细胞发生变形、坏死、空泡、萎缩;与对照组比较,中、高剂量染氟组大鼠肾脏caspase-3、caspase-9蛋白表达量[(0.37±0.05)、(0.40±0.02)和(0.41±0.07)、(0.46±0.06)]均明显增高(P<0.01)。结论氟中毒诱导大鼠肾脏组织中caspase-3和caspase-9蛋白表达增强,促进肾脏细胞凋亡。 Objective To study the effects of fluoride on the expressions of caspase-3 and caspase-9 during apopto- sis of kidney cells of rats. Methods Forty eight Sprague-Dawley (SD) rats were randomly divided into control group, low does group, medium does group, high does group, and then were given drinking-water with 0 rag/L, 50 rag/L, 100 mg/L, and 200 mg/L sodium fluoride, respectively, for 120 days. Urinary fluoride content was measured. The rats were sacrificed and their body weight and kidney organ weight were weighed. Kidney organ coefficient was calculated and renal pathological slices of the rats in high does group were observed. In high does group, caspase-3 and caspase-9 were measured with immunocytochemistry. Results The body weight of the rats in treatment groups was significantly lower than that of the control group( P 〈 0. 01 ). Kidney organ coefficient was significantly lower for the rats in medium does group than that of the control group(P 〈 0. 05). The urinary fluoride content was significantly lower in all rats with fluoride exposure than in the rats in the control group ( P 〈 0.01 ), with a dose-effect relationship between urine fluorine and fluoride concentration ( r = 0. 925, P 〈 0. 01 ). Enlarged disordered renal tubule structure, degeneration, necrosis, void and atrophy of glomerular endothelium and tubular epithelium were observed in the rats treated with fluoride. The average optical density values of caspase-3 and caspase-9 were significantly higher in the rats of medium dose fluoride group (0. 37± 0. 05 and 0.40 ± 0. 02) and high dose group (0.41 ± 0. 07 and 0.46 ± 0. 06) compared to those of the control group (P 〈 0. 01 for all). Conclusion Fluoride increases the expressions of caspase-3 and caspase-9, which induces apoptosis of kidney cells.
