摘要
目的:研究温阳解郁法对模拟早年不良经历的母婴分离小鼠的作用,探讨其相关作用机理。方法:选用从出生起连续10d、每天3h母婴分离12周-20周成年C57BL/6J小鼠,以温阳解郁复方进行4周的干预,通过旷场、高架十字迷宫及决择反应等实验观察行为模式变化;运用放射免疫法检测应激后和DexlCRH实验血清皮质酮的变化,以及用免疫组织化学方法检测中枢糖皮质激素受体(GR)和11β-羟基类固醇脱氢酶2(11β-HSD2)的表达,观察下丘脑-垂体-肾上腺皮质轴(HPA)功能状态的变化。结果:温阳解郁复方可明显增加母婴分离小鼠高架十字迷宫实验中开放臂进入次数,显著缩短趋向反应决择时间(P〈0.05),明显延长回避反应决择时间,显著降低Dex/CRH实验中血清皮质酮的高涨(P〈0.05),显著提高海马齿状回11β-HSD2的表达(P〈0.01)。结论:温阳解郁法可明显影响母婴分离小鼠行为模式和HPA轴功能状态,其主要作用位点位于海马齿状回。
Objective: To study the mechanism of maternal separation mice treated with warmtng yang wtth soothtng liver. Methods: The adult C57BL/6J mice which experienced 10 d maternal separation after birth(MS) were treated with formulae of warming yang with soothing liver for 4 weeks, then their behavior changes were observed through open field test, elevated plus maze and decision-making test, and serum corticosterone was detected by radioimmunoassay, brain glucoeorticoid receptor ( GR ) and 11β-hydroxysteroid dehydrogenase 2 ( 11β-HSD2 ) expressions by immunohistoehemistry. Results : formulae of warming yang with soothing liver markedly increased Open Arms Entries of MS mice in elevated plus maze test, significantly shortened the time of the appetitive response ( P 〈 0.05 ) , and obviously extended the time of avoidance response in decision-making test, significantly reduced serum corticosterone in Dex/ CRH test (P 〈 0. 05) and increased 11β-HSD2 expression in dentate gyrus (P 〈 0.01 ). Conclusion: warming yang with soothing live has significant effects on behavior pattern and hypothalamic-pituitary-adrenocortical axis (HPA) function in MS mice, its target site may be in hippoeampus dentate gyrus.
