重组人促红细胞生成素后处理对肢体缺血再灌注诱发大鼠肾损伤的影响 被引量:1

摘要 目的探讨重组人促红细胞生成素(rHu-EPO)后处理对肢体缺血再灌注(IR)诱发大鼠肾损伤的影响,以及核转录因子-κB亚基p65亲和肽(NF-κB p65)、TNF-α在其中的作用。方法选择健康成年雄性SD大鼠30只,随机分为假手术组、肢体IR组、rHu-EPO后处理组(rHu-EPO组)各10只。3组均于再灌注2 h后检测血清尿素氮(BUN)、肌酐(Cr),检测肾组织中的超氧化物歧化酶(SOD)活性、丙二醛(MDA)、TNF-α及NF-κB p65表达。结果与假手术组比较,肢体IR组、rHu-EPO组的血清BUN与Cr、肾组织中的MDA与TNF-α升高,SOD活性降低(P均<0.05);与肢体IR组比较,rHu-EPO组BUN、Cr降低,肾组织中的MDA、TNF-α降低,SOD活性升高(P均<0.05)。肢体IR组肾组织中的NF-κB p65阳性表达明显,rHu-EPO组的NF-κB p65阳性表达高于假手术组、但低于肢体IR组(P均<0.05)。结论 rHu-EPO可减少氧自由基、TNF-α生成,抑制NF-κB p65表达,对大鼠肢体IR诱发的肾损伤有保护作用。
