摘要
目的了解2009年--2011年大连地区肠道病毒71型(EV71)分离株的基因特征,初步探讨VPl、VP4、5’UTR区核苷酸和氨基酸序列变异与不同临床类型的关系。方法从手足口病例、重症病例的粪便标本中分离获得9株EV71病毒株,RT—PCR扩增VPl、VP4、5’UTR区,并进行序列测定和分析。结果这9株EV71毒株均属于C4a亚型,其核苷酸之间的同源性为88.7%~99.9%;氨基酸之间的同源性为96.7%~100%;9株毒株的VP4氨基酸序列高度一致;对于5’UTR区在第208位核苷酸,重症来源的样品发生了碱基置换(G—A);在第254位核苷酸,重症来源的样品发生了碱基置换(A—G)。氨基酸序列未发现一致性改变。结论(1)2009年--2011年大连地区分离的9株EV7I毒株均属于C4a亚型。(2)基于VP4的基因分型与VPl的基因分型相一致,均为C4a亚型,且VP4序列高度保守。(3)在5’UTR序列上发现了第208位和254位核苷酸的碱基置换,这一结果还有待于进行下一步的研究。
Objective To understand the genetic characteristics of enterovirus 71 isolates in Dalian from 2009 to 2011, and to discuss the relationship between variations of VP1, VP4, 5'UTR gene an fferent clinical types. Methods The VP1, VP4, 5'UTR genes of 9 enterovirus 71 isolates from stool specimens of hand -foot -and mouth disease cases were amplified with re- verse transcription -PCR( RT- PCR) and their sequences were analyzed. Results The 9 enterovirus 71 isolates belonged to C4a subtype, and the nucleotide homology was between 88.7% and 99.9% ; while that of amino acid was between 96.7% and 100%. The VP4 region sequence analysis showed the 9 isolates were highly consistent. For 5'UTR region, base substitution ( G →A) was found in the 208th nucleotides of severe HFMD cases, and base substitution (A→G) was found in the 254th nucleo- tides of severe HFMD cases. The amino acid sequence had no consistent change. Conclusion Nine enterovirus 71 isolates in Dalian from 2009 to 2011 belonged to the FAa subtype. Genotyping based on VP4 was consistent with that based on VPI, all belonged to C4a subtype, and the VP4 sequence was highly conserved. The 5'UTR sequence was found base substitution on the 208th and 254th nucleotide, needing future research.
