摘要
目的观察胰淀素对体外培养胰岛细胞活力的影响,探讨胰淀素在2型糖尿病发病中的作用。方法应用体外单 层培养新生乳鼠胰岛细胞,采用噻唑蓝法测定在不同浓度胰淀素作用不同时间后细胞的活力变化。结果10 μmol/L以 上胰淀素能引起胰岛细胞活力降低(P<0.01),10 μmol/L胰淀素时间梯度组细胞活力呈进行性下降,在 2 h时与对照组 相比差异非常显著。相差显微镜下可见部分细胞胞体缩小,胞质发生空泡变性,少数细胞核固缩、裂解,胞膜突起呈出胞 状。结论 胰淀素对胰岛细胞有直接损伤作用,这种损伤程度与胰淀素浓度及作用时间呈正相关。
Qbjective To investigate the effect of amylin on the activity of pancreatic islet cells, and to explore its role in the occurrence of type 2 diabetes mellitus. Methods pancreatic islet cells from neonatal rats were cultured in vitro, the activity of which was determined by MTT assay after incubation with amylin at different concentrations for varied period of time. Results Theactivity and proliferation of the islet cells were significantly reduced when amylin concentration was higher than 10 μmol/L (P<0.01). When incubated with 10 μmol/L amylin for different time, the islet cells showed increasingly inhibited viability and proliferation by amylin with the prolongtion of incubation time; such effect of amylin was most obvious at 2 h when compared with the control group. Shrinkage, vacuolar degeneration of the cytoplasm were observed in some cells, and a few cells presented nuclear pyknosis, karyorrhexis and processes of the cellular membrane. Conclusion Amylin acts to directly damage islet cells through decreasing the viability and proliferation of the cells, and the degree of the damage was positively correlated to the concentration and treatment time of amylin.
