摘要
采用环介导等温扩增技术(PMA-LAMP)对克氏螯虾(Cambarus Clarkii)样品中沙门氏菌进行快速检测,旨在建立一种快速简便、灵敏度高、特异性强的检验方法。分别采用PMA-LAMP、PCR检测方法对沙门氏菌进行检测。结果表明,PMA-LAMP法检测限为2×10 CFU/mL,PCR法和GB/T 4789.4-2008检测限为2×103CFU/mL。采集湖北省省内60份市售的克氏螯虾样品,分别采用PMA-LAMP检测方法、PCR检测方法及国家标准方法(GB/T 4789.4-2008)对样品进行检测,PMA-LAMP方法检出6例沙门氏菌,阳性率为10%;PCR检测方法检出4例沙门氏菌,阳性率为6.7%;国家标准方法检出4例沙门氏菌,阳性率为6.7%。
Loop-mediated isothermal amplification for rapid detection of Salmonella in samples of crayfish (Cambarus clarkii)was develeped to provide a quick and easy,highly sensitive and specific testing methods.PMA Loop-mediated isothermal amplification detection method,PCR detection method and national standard(GB/T 4789.4-2008) were used to isolate and identification of Salmonella.The results showed that the detection limit of the PMA-LAMP assay was 2×10 CFU/mL,indicating the sensitivity of PCR was 2×103 CFU/mL.PMA-LAMP method,PCR method and national standard (GB/T 4789.4-2008) were used to detect crayfish samples.PMA-LAMP method detected six cases with the positive rate of 10%.PCR method detected four cases with the positive rate of 6.7%.National standard method detected four case with the positive rate of 6.7%.
