摘要
目的 研究逆转录酶抑制剂 (AZT)对卵巢肿瘤细胞端粒酶活性的影响 .方法 采用 TRAP- EL ISA方法检测卵巢癌 HO- 8910细胞在 AZT作用前后端粒酶活性的变化 ,以流式细胞仪分析细胞周期的改变 .结果 AZT作用后的 HO-8910细胞端粒酶活性明显受抑制 ,且细胞的 G2 /M期 DNA含量明显增高 .结论 AZT能明显抑制肿瘤细胞的端粒酶活性 .
AIM To study the effect of inhibition of telomerase activity by reverse transcriptase inhibitors (AZT) on ovarian cancer cells in vitro. METHODS Telomerase activity was determined by TRAP-ELISA in untreated and treated HO-8910 cells by AZT, cell cycle phases were analyzed by flow-cytometry. RESULTS Telomerase activity of HO-8910 cells was significantly inhibited when treated with AZT, and cell cycle of treated HO-8910 cells was changed with marked increase in G 2/M phase. CONCLUSION AZT can effectively inhibit telomerase activity of tumor cells, and represent an effective and novel approach for treatment of cancer.
