摘要
目的 改进RP-HPLC法测定牛黄及安宫牛黄丸中的游离胆红素以保证牛黄类药材及安宫牛黄丸的质量与用药安全.方法 以二氯甲烷为溶剂提取样品,HPLC法测定游离胆红素的量.对于安宫牛黄丸,分别考察碳酸钙、硅藻土等不同分散介质对游离胆红素测定的影响.色谱条件为Kromasil 100(A) C18色谱柱(5μm,0.46 cm× 15cm),以乙腈-1%冰醋酸(95∶5)为流动相,检测波长为450 nm.结果 胆红素在0.305 5~ 30.55μg/mL范围内线性良好,加样回收率在95% ~ 105%之间.对于安宫牛黄丸,采用碳酸钙作为分散介质的测定结果优于硅藻土等介质.结论 采用碳酸钙替代硅藻土作为牛黄类药材及其大蜜丸样品中游离胆红素分析的分散介质,方法准确度高,操作简便易行.
AIM To use RP-HPLC modification to improve the method for unconjugated bilirubin determina- tion in bezoar and Angong Niuhuang Pills in order to assure their quality and safety. METHODS Unconjugated bilirubin in bezoar and Angong Niuhuang Pills was eluted on a Kromasil Cls column (5 μm, 0. 46 cm × 15 cm) by an ioscratic mobile phase composed of acetonitrile-l% acetic acid (95 : 5) and detected at 450 nm. For Angong Niuhuang Pills, different kinds of dispersion medium such as CaCO3 , diatomite were investigated. RESULTS Unconjugated bilirubin was well detected in samples and its linear range was 0. 305 5 -30. 55 μg/mL with average recoveries of 95% - 105% . CONCLUSION The experiment shows that method of CaCO3 replacing diatomite as dispersion medium in honey bolus is easy to determine the contests of unconjugated bilirub in bezoar and Angong Niuhuang Pills with high accuracy.
