摘要
目的 建立测定人血浆中拉莫三嗪浓度的高效液相色谱法。方法 色谱柱为SunFire C18(4.6 mm×250mm,5μm),流动相为水-乙腈-三乙胺(64.5:35:0.5,v/v/v),磷酸调节pH至6.5,流速为1.0 mL min-1,血浆标本经乙酸乙酯-二氯甲烷(75:25,v/v)提取后,在237 nm波长下进行检测。结果 拉莫三嗪在0.312 5~40.0μg mL-1与峰面积呈良好的线性关系(Y=0.047 2X+0.069 3,r2=0.999 2),相对回收率均在82%~105%,日内和日间RSD均〈5%。结论 该方法专属性强、准确度好、灵敏度高、操作简便,适合临床上对拉莫三嗪血药浓度的常规监测。
Objective To establish a high-performance liquid chromatogramphy to determine lamotrigine in the plasma. Methods The analytical column was SunFire C18(4.6 mm×250 mm, 5 μm). The mobile phase was composed of water-acetonitrile- triethylamine(64.5 :35 :0.5, v/v/v, pH = 6.5) at 1.0 mL · min- 1. The detection wavelength was 237 nm. Plasma samples were extracted with ethylacetate-dichloromethane(75 :25, v/v). Results The lamotrigine had a good linearity at 0.312 5- 40.0 μg · mL- 1(Y = 0.047 2X + 0.069 3, r2= 0.999 2), and the relative recovery was 82%- 105%. Both the interday and intraday relative standard deviations were less than 5%. Conclusion The method is rapid, sensitive and suitable for clinical therapeutic drug monitoring.
