穿心莲内酯对白假丝酵母菌菌丝的影响

The effects of andrographolide on hyphal development of Candida albicans

摘要 目的探讨中药有效成分穿心莲内酯(Andrographolide,AG)对白假丝酵母菌菌丝的影响。方法利用稀释涂布法观察AG对固体培养基上白假丝酵母菌菌落形态的影响;倒置显微镜观察AG对白假丝酵母菌芽管及菌丝形态的影响;采用XTT还原法评价AG对白假丝酵母菌菌丝活性的影响;采用实时荧光定量PCR(qRT-PCR)检测白假丝酵母菌菌丝形成相关基因SUN41、CSH1以及CaPDE2表达量变化。结果 AG能影响白假丝酵母菌菌落的形态;倒置显微镜观察表明AG能够抑制白假丝酵母菌菌丝及芽管的形成;qRT-PCR检测显示AG能使SUN41和CSH1基因表达下调和使CaPDE2上调。结论穿心莲内酯可能通过影响SUN41、CSH1与CaPDE2基因的表达而抑制白假丝酵母菌菌丝的形成。 Objective To investigate the effect on the hyphal development and the morphology of colony by an- drographolide ( AG), an effective component of Traditional Chinese medicine. Methods XTY assay was used to evaluate the effect of AG( 1 000 μg/mL, 500 μg/mL and 250 μg/mL) against the attachment of Candida albi- cans ; Inverted microscope was applied to observe the morphological changes of hyphal development treated by AG; The real-time fluorescence quantification PCR (qRT-PCR) was adopted to inspect expression of hyphal develop- ment-related genes, including SUN41, CSH1 and CaPDE2. Results AG could obviously affect Candida albicans during the hyphal deve!opment, and the expressions of SUN41 and CSH1 were down-regulated, CaPDE2 up-regu- lated. Conclusion AG can inhibit hyphal development of Candida albicans by regulating the expressions of SUN41, CSH1 and CSH genes.
