摘要
目的建立茯苓皮中松苓新酸的HPLC测定方法。方法采用高效液相色谱法,Lichrospher C18色谱柱(200 mm×4.6mm,5μm),流动相为乙腈–0.5%磷酸水溶液(80∶20),检测波长为242 nm,体积流量为1.0 mL/min,柱温为30℃,进样量为10μL。结果松苓新酸在0.142 5~2.280 0μg与峰面积呈良好的线性关系(r=0.999 3),平均回收率为101.83%,RSD值为1.45%(n=6)。结论该方法操作简便,稳定可行,重复性好,可用于茯苓皮中松苓新酸的测定。
Objective To establish an HPLC method for the determination of dehydrotrametenolic acid in the peels of Poria cocos. Methods HPLC was carried out on a Lichrospher C18 column (200 mm×4.6 mm, 5 μm) with acetonitrile-0.5% phosphoric acid (80 : 20) as mobile phase. The detection wavelength was set at 242 nm. The injection volume was 10 gL at the flow rate of 1.0 mL/min. The column temperature was at 30 ℃. Results The calibration curve was linear over the range of 0.142 5--2.280 0μg (r=0.999 3) for dehydrotrametenoli6 acid. The average recovery rate was 101.83% with RSD value at 1.45% (n=6). Conclusion The established method is rapid, sensitive', and accurate, and could be used for the determination of dehydrotrametenolic acid in the peels of e. cocos.
