摘要
旨在运用半定量RT-PCR技术探究不同浓度盐胁迫下3种纤维亚麻耐盐基因NHX的表达规律。提取不同浓度盐胁迫6天的3种亚麻无菌苗茎叶组织总RNA,反转录得到cDNA,利用优化的半定量RT-PCR法检测3种亚麻无菌苗的NHX基因表达量。结果表明,‘范妮’和‘双亚5号’无菌苗经100 mmol/L NaCl胁迫6天后NHX表达量最高,‘天鑫3号’无菌苗经200 mmol/L NaCl胁迫6天后NHX基因表达量最高。与对照相比,3种纤维亚麻经盐胁迫后NHX基因表达量均上调,因此可将NHX基因的检测作为鉴定这3种纤维亚麻耐盐性的依据。
The objective was to explore salt tolerance gene NHX expression pattern in three kinds of fiber flax using semi-quantitative RT-PCR technology stressed with NaC1. Total RNAs were extracted respectively from the sterile stem-leaf tissues stressed with different concentration of NaC1 for 6 d. eDNA were reversely transcribed. Expression quantities of NHX gene in the different groups were detected by optimized semi-quantitative RT-PCR method. The results showed that the highest expression quantities of NHX gene in ' Fanny' and 'Shuangya 5' sterile seedlings stressed with 100 mmol/L NaC1 for 6 d and in ' Tianxin 3' sterile seedlings stressed with 200 mmol/L NaC1 for 6 d. Expression quantities of NHX gene in the three kinds of fiber flax after salt stresses were all up regulation compared with contrasts. Therefore, detection of NHX gene could be used as an index judging salt tolerance of these three kinds of fiber flax.
