摘要
为研究继代次数对除虫菊组培苗生长状态及其除虫菊酯质量分数的影响,采用高效液相色谱法测定了除虫菊‘F1-10-4’基因型组培苗不同继代次数叶片中除虫菊酯质量分数的变化,并统计分析了组培苗生长状况。结果表明,经过5次继代除虫菊叶片中除虫菊酯质量分数呈先上升再下降的趋势,其中第3次继代时达到最大值1.997 mg/g。同时,继代次数对除虫菊组培苗生长表现也产生了一定的影响,其中,第3代组培苗根系粗壮、主枝发达、叶片深绿,总体植株生长健壮;而第五代组培苗表现出枝叶纤细、生长缓慢和植株矮化的现象。由此认为,在继代3次时除虫菊基因型‘F1-10-4’组培苗整体长势最好,除虫菊酯质量分数最高,更多次继代除虫菊组培苗生长不佳,也增加组培成本。
For the study of subculture times on the growth and pyrethrins mass fraction of pyrethrum,we used high performance liquid chromatography (HPLC) method to determine the content changes of pyrethrins,and at the same time thc growth vigor of pyrethrum plantlets in vitro was also analyzed.The results showed that with subculture for 5 times the content of pyrethrins increased first and then decreased,on the 3rd times of subculture reached the highest 1.997 mg/g.Mcanwhile,the times of subculture had impact on the vigor and roots growth of plantlets.The plantlets at the 3rd times of subculture were more vigorous with stronger roots than at the 1st times of subculture.At the 5th times of subculture,the plantlets showed slender foliage,slower growth phenomenon than other 4 times of subculture.Thus,the leaves of pyrethrins content of 'F1-10-4' genotype had the strongest growth vigor at the 3rd times of subculture.Further subculture time did not improve leaves' pyrethrins contents and growth of pyrethrum and it could increase the cost.
